Content Determination

Preparation of reference solution: take a proper amount of Ruscogenin reference solution, accurately weigh it, and add methanol to make it containing 50% per 1ml μ G solution.Preparation of standard curve accurately measure 1 ml, 2 ml, 3 ml, 4 ml, 5 ml and 6 ml of the reference solution, place them in a conical flask with a stopper respectively, and evaporate the solvent in a water bath. Precisely add 10ml perchloric acid, shake it well, keep it in hot water for 15 minutes, take it out, cool it with ice water, take the corresponding reagent as blank, measure the absorbance at the wavelength of 397nm according to ultraviolet visible spectrophotometry (Appendix V A), take the absorbance as the ordinate and the concentration as the abscissa, and draw the standard curve.

Preparation of test solution: take about 3G fine powder of the product, accurately weigh it, place it in a conical flask with a stopper, accurately add 50ml of methanol, weigh it, heat and reflux for 2 hours, cool it, weigh it, make up the lost weight with methanol, shake it well and filter it. Accurately measure 25ml of continuous filtrate, put it into a flask, recover the solvent to dryness, add 10ml of water to dissolve the residue, saturate it with water, shake it with n-butanol for 5 times, 10ml each time, combine the n-butanol solution, wash it twice with ammonia test solution, 5ml each time, discard the ammonia solution, and evaporate the n-butanol solution to dryness. Dissolve the residue with 80% methanol and transfer it to a 50ml volumetric flask, add 80% methanol to the scale and shake well.

Determination method accurately measure 2 ~ 5ml of the test solution, place it in 10ml plugged dry test tube, according to the method under the preparation of the standard curve, measure the absorbance according to law from "volatilizing the solvent in the water bath", read out the amount of ruscoegenin in the test solution from the standard curve and calculate it.

The total saponins of Ophiopogon japonicus shall not be less than 0.12% based on Ruscogenin (C27H42O4).
